Lesson #3 of the RxeSearch curriculum involves exploring a hypothetical enzyme assay found in a pathogenic microorganism.  The kit provides the replacement reagents for the basic assay and screening the chemical library.  Each kit includes the following materials:

1 Bottle of Substrate (60 ml of 50 mM lactic acid)

1 Bottle of Buffer (60 ml of 200 mM Tris, pH 8)

Investigating Enzymes Kit

Examine the effects of pH, temperature, and inhibitors on enzymes.

Investigating Enzymes provides the materials for 15 student groups to run enzyme assays and examine the parameters that affect activity.  This kit uses lactate dehydrogenase which can be measured by the conversion of  lactic acid to pyruvate.  The NAD dependent reaction generates a red color that can be observed visually qualitatively or quantitatively using a spectrophotometer.  The kit comes with five buffers (pH 5-9) which are used to demonstrate the impact of pH on enzymes.  A set of inhibitors is also included.  Guidelines for temperature effects and temperature of inactivation are discussed in the teacher's guide.  Each kit includes:
